Multiple Grid Staining Device

Instruction for use:
- Insert grids edgewise
in the letter coded disc (D). The disc is divided into two equal sections
identified by I and II scribed in the plastic. (Figure 1)
- Press the fine nickel mesh disc (M) into the cup (C).
It prevents the grids from dropping out through the openings in the
cup.
- Remove the lid (L) from the perfusion chamber.
- Place the cup (C) over the coded disc (D). Holding the
assembly together, invert it and place it in the perfusion chamber
(P). (Figure 2)
- Replace lid (L) and tighten until the thin dark band appears at the
'O' ring. This indicates 360°Contact
and a tight seal on the 'O' ring.
- Insert tygon tubing from elevated rinse source into larger
side orifice. (Make sure tube is clamped shut and fluid has been
drawn to point of clamping)
- Insert tygon tubing from lower waste fluid container into
top orifice.
Staining Procedure:
Note: All stains entering chamber can be filtered. Lead stains should be centrifuged, then filtered.
- Using regular syringe needle with luer-lock fitting, draw
8 ml of stain solution into body of syringe.
- Place clean filter in Swinny filter holder.
- Replace needle with Swinny filter holder.
- Insert syringe into remaining side hole on strainer.
- Inject stain slowly. Be sure no air bubbles are trapped
in grid pockets. If so, tap sides of grid stainer to shake loose.
Tip stainer to remove all through drain tube. Stain should extend into
drain tube.
- Stain for desired time.
- Release rinse clamp. Rinse for desired duration. (Rinse
rate through tubing is approximately 1 ml/second.)
- Replace clamp on rinse line. While stainer is still inverted,
remove syringe. Allow residual rinse to drain (shake once or twice
to facilitate draining.)
- Repeat entire procedure with the next stain, if desired.
- Disassemble and remove cup. Press cup down on absorbent
paper. Dabpaper on exposed surface of grid disc.
Note: One may wish to vacuum dry disc and grids.
- Cover grid holder to exclude dust or remove grids and
store in desired manner.
Note: For more convenience, you may substitute
Swinny (step 2 and 3 above) by Syringe Filter Disc, EMS Cat #67002 for
13mm Membrane diameter, 0.22 m pore size)
Cleaning Procedure:
Add a small amount of mild detergent* to distilled water. Wash ultrasonically for one hour. Rinse thoroughly for ten minutes in distilled water. Air dry in dust-free environment.
Note : Strong detergent or solvent will degrade plastic.
* We recommend using EMS 'Micro
Cleaning Solution' Cat. #71054, 1 qt bottle.
