Van-Gieson's Method for Collagen Fibers
EMS Catalog #: 26350-Series
Fixation:
10% Buffered Neutral Formalin (#15740) or fixative of choice.
Sections:
Paraffin at 6µm or Celloidin.
Staining Procedures:
- Deparaffinize and hydrate to distilled water.
- Weigert’s Working Hematoxylin* for 10 minutes)
- *Weigert’s Working Solution is prepared by mixing equal parts of Weigert’s Iron Hematoxylin A (#26350-02) and Weigert’s Iron Hematoxylin B (#26350-03)
- Wash in distilled water.
- Stain 1 – 3 minutes in Van Gieson’s Solution (#26350-01)
- Dehydrate in 95% alcohol, absolute alcohol, two changes each. Clear in 2 changes Xylene (#23400).
- Mount with Permount (#17986-01) or DPX (#13510).
Stain Results:
| Collagen | Red |
| Smooth and striated muscle | Yellowish to brownish |
| Cornified epithelium | Yellow |
| Hyalin | Yellow |
| Nuclei | Blue to Black |
