Prussian Blue Method for Hemosiderin

Fixation:

10% Buffered Neutral Formalin (#15740).

Sections:

Paraffin @ 5 microns

Staining Procedures:   

  1. Deparaffinize and hydrate to distilled water.
  2. Place sections in working staining solution* for 30 minutes @ 60°C or for 1 hour at room temperature. Rinse in distilled water.
    1. Prepare working stain by mixing equal volumes of Potassium Ferrocyanide, 2% Aqueous (#26613-01) and Hydrochoric Acid, 2% (#26613-02). Prepare fresh just before use.
  3. Counter-stain in Safranin O, 0.2% (#26613-03) for 2 minutes. Wash in Acetic Acid, 1% (#26613-04)
  4. Dehydrate in 95% Alcohol, absolute alcohol, and clear in xylene, two changes each.
  5. Mount with Permount (#17986-01) or any synthetic resin.

Stain Results:  

Hemosiderin Blue or Green
Nuclei Red
Background Pink
