Adherent Cells HEK293 (Human Embryonic Kidney) Initial Observation: Settled equally well onto all plates 1hr after seeding (25% confluence) After 72 Hours: Cells have grown to 80% confluence Detachment: Some cells detach from the bottom by tapping plates. Incubation with trypsin allows for easy detachment of all cells for a first passage into new plates. After 72 Hours: Cells have again grown to confluency.
COS (Fibroblast-like cells) Initial Observation: Settled equally well onto all plates 1hr after seeding. Patches of cells are larger patches than HEK cells. After 72 Hours: Cells are 100% confluent, media is noticeably more acidic (by colorimetric pH indicator) than on plates containing other cell types, indicating overgrowth. Detachment: Cells are difficult to detach from plates. After incubation with trypsin and vigorous resuspension cells are successfully passaged at a lower seeding density to prevent overgrowth. After 72 Hours: Cells are again close to 100% confluent, media does not appear overly acidic.
Suspension Cells HEK293 (Human Embryonic Kidney) Initial Observation: Nearly identical to adherent cells in growth rate and appearance on plates. Detachment: Easy to detach without trypsin, but trypsin incubation is still more efficient than tapping alone. After 72 Hours: Cells are approximately 60% confluent, some patches floating in the media
Conclusion There is little difference in growth rate/percentage confluence and the adhesion/ease of detachment as compared to control dishes.
