Gomoris’ Methenamine Silver for Urate Crystals

Fixation:

Absolute Alcohol at 4°C for overnight.

Sections:

Paraffin @ 4 - 5 microns

Staining Procedures:   

  1. Deparaffinize to Xylene and rinse with 3 changes of absolute alcohol.
  2. Place slides in preheated working* Methenamine Silver. Incubate for 30 minutes @60°C.

To prepare working solution:

Methenamine-Silver Nitrate Stock Solution:

Silver Nitrate, 5% (#26614-01) - 5 ml
Methenamine, 3% (#26614-02) - 100 ml

Methenamine-Silver Nitrate Working Solution:

Methenamine Silver-Nitrate Stock Solution: - 25 ml
Distilled water: - 25 ml
Sodium Borate, 5% (#26614-03): - 2 ml
Prepare fresh prior to use. DO NOT USE IF CLOUDY. Urate Crystals should be black

  1. Rinse sections in distilled water.
  2. Tone with Gold Chloride, 0.1% (#26614-04) for 5 minutes.
  3. Give sections 4 or 5 rinses of distilled water.
  4. Sodium Thiosulfate, 3% (#26614-05) for 5 minutes.
  5. Wash in tap water for 5 minutes.
  6. Rinse in distilled water.
  7. Counterstain in Light Green Working Solution (#26614-06) for 2 minutes.
  8. Dehydrate, clear and  mount with Permount (#17986-01) or any synthetic resin

Stain Results:  

Urate Crystals Black
Background Green
