Harris Hematoxylin (With or Without Mercury)

Fixation:

Formalin 10% Neutral Buffered (#15740) or fixative of choice

Sections:

Paraffin @ 5 -6 microns

Staining Procedures:

  1. Deparaffinize and hydrate to distilled water
    1. Stain in Harris Hematoxylin (#26754-01) containing 2 – 4 ml Glacial Acetic Acid per 100 ml for 5 – 15 minutes. Wash in water.
    2. If using Harris Hematoxylin w/o Mercury (#26754-1A) containing 2 – 4 ml Glacial Acetic Acid per 100 ml for 5 – 15 minutes. Wash in water.
  2. Differentiate in Acid Alcohol (#26754-03) until the sections turn red.
  3. Blue section in Sodium Acetate, 1% (#26754-04) or Ammonium Water (#26754-4A) or Lithium Carbonate Solution 26754-4B).
  4. Wash thoroughly and counterstain.
